Generally, it would be possible to scan the device without opening. Check out what your AV program can do. What are you using for AV? I looked at Windows Defender; looks like it will scan all locations unless some are excluded. I'd want to be sure though.

Just open up defender by going to charms, search, type in defender, select defender, select settings tab, then go to the advanced tab on left hand side, tick box to "scan removable drives", save changes...insert your usb drive and run defender.

Not sure what it is. Haven't experienced it. Can you scan it specifically in Kaspersky. Try it, but be sure all is OK before letting anything on it loose.

First you need to turn off Autorun. Autorun is a Windows feature that automatically executes using what instructed in a hidden file called Autorun.inf resided in the root folder of your Drive(s) and hackers use this to spread viruses to your PC.

Win+X -> Run
and type gpedit.msc

Administrative Templates >Windows Components > Autoplay Policies
, double click "Turn off Autoplay", Next is click Enabled, and then select All drives so that you can disable Autorun on all drives.

Next, Set your folder Options to show Hidden File then plug in your USB stick, look in the root folder, if there is an Autorun.inf , edit it with notepad to find out what is being instructed to run then delete it, if there's an executable file or a vbscript etc.., you will need to delete those also . In addition, check the root folder of all Drives connected to your PC.

To learn more about autorun, Please read this article:
Is autorun really that evil, and if so, how do I turn autorun off?
