I've also been getting the same error code with Defender, so I tried what you suggested. Upon completion of the scan through sfc/scannow, Command Prompt game me a message saying that it'd found some corrupt files but that it was unable to fix some of them. What do I do now?

By the way, I also get Error Code 0x80040154 when I try to do an Elevated activation of Windows Defender with Admin privileges. It also gives me an error message saying, "Class not registered".

Also, even though Defender's Real-time protection and Firewall are on, and it's showing that it's up-to-date (unless I attempt definition updates through Defender rather than Windows Update), Action Center still has two messages all the time right now, talking about antivirus and antispyware software. How do I fix these problems?
