It's definitely not a hardware problem as one minute it's all working, then it suddenly stops. I've tried everything. My sound card drivers are all up to date, it also happens at the same time as youtube videos stop playing on google chrome (despite them loading), windows media player is unable to play and an error occured on youtube on firefox.
However after a few hours it finally lets me on the "configure speakers" setting and I click test and it begins to play sounds again, from then on it plays sound and loads videos. Is this a virus or a software problem? Please help!

Edit: When I click "test" it tells me "The device is being used by another application. Please close any devices that are playing audio to this device and then try again."

You did not indicate you computer make and model also the sound card.

Uninstall the sound card driver in Device Manager and reboot, Windows will install its generic driver and check if the sound returns to normal.

Have you ran any registry cleaner lately, something may have corrupted Windows and a Refresh may be in order.

You may also try sfc /scannow in the elevated command prompt, it will take a while.

Problems with audio update KB2962407

After a recent Windows 8 Update (July 2014) some computers lost audio completely, forcing the user to perform several unsuccessful tests, including:
enter properties of the speakers, run the sound test, getting the following message: "NO OTHER APPLICATION CLOSING DEVICE USING ALL DEVICES THAT ARE PLAYING AUDIO AND TRY AGAIN."
or:
entering the device manager, and sound and video controllers and game, disabling the audio driver.
But after a while the problem continued, there was no audio again! . Users of the Microsoft forums JUANGIMENE gave us the answer:
OPEN PANEL CONTROL ... AND CLICK uninstall PROGRAMS AND WHEN OPENING AT THE TOP OF THE SCREEN WHERE IT SAYS SHOW UPDATES select WHICH WILL OPEN WITH ALL YOU HAVE AND SEEK THE WINDOWS UPDATE KB2962407 SAYING THIS AND CLICK uninstall restart PC AND SOLVED ...
(All other recommendations were only temporary, eye)
The problem is the update KB2962407! as discussed in the following forum:
